Precise microwave heating has emerged as a very valuable tool in SPPS. Biotage® Initiator+ Alstra™ is a single-channel robotic peptide synthesizer with microwave heating. Here we describe the use of the Initiator+ Alstra for the synthesis of both ‘difficult’ sequences, such as the Jung&Redemann peptide, as well as for the synthesis of long sequences. We demonstrate the flexibility of the instrument and the use of the oscillating mixer to enable syntheses at a 0.1 mmol scale in the small reaction vessel thereby allowing the efficient incorporation of high-value building blocks.
